Tessamorelin is a synthetic growth hormone–releasing hormone (GHRH) analogue designed for enhanced stability and potency compared with native GHRH. Preclinical pharmacology shows that it stimulates pituitary somatotrophs via GHRH receptor activation, leading to increased pulsatile GH release and downstream IGF-1 signaling in animal models. Studies in vitro and in vivo have examined its receptor-binding affinity, peptide stability, and endocrine cascade activation, as well as gene-expression changes associated with GH-dependent tissue responses. The literature emphasizes its utility in probing GHRH receptor biology and the regulation of the somatotropic axis in nonclinical systems.
